Challen) saat tengah merundingkan bisnis dengan teman-temannya. Alice terbangun dari tidurnya. Ia lalu bercerita pada ayahnya tentang mimpinya yang aneh. Ia jatuh ke lubang dan bertemu kelinci berjas, kucing yang tersenyum dan ulat berwarna biru. Snene lalu beralih ke perjalanan Alice dewasa (Mia Wasikowska) dengan kereta kuda ke tempat pesta pertunangannya berlangsung. Alice ditunangkan pada Hamish (Leo Bill), tapi ia malah lebih tertarik pada kelinci berjas yang terus ia impikan sejak masih kecil. Karena itu saat Hamish melamarnya di depan semua tamu yang hadir, Alice berujar ia perlu waktu dan berlari mengejar kelinci ke sebuah pohon. Alice jatuh ke dalam lubang, persis seperti mimpinya. Alice lalu meminum cairan dalam botol yang ia temukan sehingga tubuhnya menyusut dan bisa masuk melalui pintu. Ia lalu memakan makanan yang ia temukan dan tubuhnya menjadi berukuran raksasa. Ia sampai di sebuah taman dan bertemu dengan Tweedledee, Tweedledum (Matt Lucas), White Rabbit (Michael Sheen), dan Dormouse (Barbara Windsor). Mereka memberitahu Alice bahwa menurut ramalan oracle, Alice adalah orang yang diramalkan akan mengalahkan Jabberwocky (Christopher Lee). Mereka membawa Alice kepada Absolom, ulat bulu berwarna biru yang senang merokok untuk menanyakan apakah Alice Kingsleigh memang orang yang selama ini mereka nantikan untuk memenuhi ramalan. Belum selesai bicara, tiba-tiba mereka dikepung oleh pasukan Red Queen dan Alice dikejar Bandersnatch. Ia lalu bertemu dengan Cheshire Cat (Stephen Fry) yang lalu mengobati luka Alice akibat ulah Bandersnatch dan Mad Hatter (Johnny Depp) yang menyembunyikan Alice di dalam poci teh agar tidak ditangkap Ilosovic Steyne, kaki tangan Red Queen (Helena Bonham Carter) yang mencari Alice. Bersama Mad Hatter dan White Rabbit, Alice lalu menyusup ke istana untuk mencari pedang vorpal untuk membunuh Jabberwocky. Sayangnya misi mereka gagal dan Mad Hatter dipenjara karena menolak memberitahu di mana keberadaan Alice. Red Queen yang tidak mengetahui identitas Alice menerimanya tinggal di dalam istananya. Alice lalu mengembalikan bola mata Bandersnatch sehingga Bandersnatch jadi berpihak padanya dan membiarkan Alice mengambil pedang vorpal. Bahkan Bandersnatch yang menyelamatkan Alice saat identitas Alice terbongkar saat mencoba menyelamatkan Mad Hatter. Alice lalu menemui White Queen. Berkat pertolongan Cheshire Cat, Mad Hatter juga berhasil bebas dari hukuman mati dan menyusul Alice. Meski sempat ragu, Alice akhirnya setuju untuk mewakili White Queen melawan Jabberwocky, sementara Mad Hatter mengalahkan Ilosovic. Red Queen dan Ilosovic akhirnya ditangkap dan takhta kerajaan diberikan pada White Queen. Alice pun memutuskan pulang meski sempat diminta tinggal oleh Mad Hatter. Alice berjanji akan kembali ke Wonderland. Ia lalu merangkak keluar dari lubang dan kembali pada saat pesta pertunangannya. Ia menolak lamaran Hamish dan memilih berlayar ke China untuk meneruskan cita-cita ayahnya berbisnis hingga ke penjuru dunia.
