Total aerobic microbial count (plate method:(
  N.B;The limit of m.o in herbal pharmaceutical preparations should not exceed 10,000 / gm of plantmaterial.1-Suspend 10.0 g(solid) or 10.0 ml(liquid) of the specimen influid soybean casein digest medium(in PH7.2 phosphate buffer) to make 100 ml.  N.B;Add the specimen to medium not more than 1 hour after preparing the appropriate dilutions for inoculation.2-Pipet 1 ml of the final dilution onto each of two sterile Petri dishes. 3-Promptly add to each dish 15 to 20 ml of soybean casein-digest agar mediumthat previously has  been melted and cooled approximately 45 °C.4-Cover the Petri dishes, mix the sample with the agar by tilting or rotating the dishes, and allowthe contents solidify at room temperature.5-Invert the Petri dishes and incubate for 48 to 72 hours. 6-after incubation period examine the plates growth, count the number of colonies, and express theaverage for two plates in terms of # of m.o / g or ml of specimen.Example;If the average number of colonies in both Petri dishes = 190.The result will be 190 X 10 = 1.9 X 10² / g or ml.  N.B;1-We here make dilution of 1:10 which we expect to give colonies number between 30 and 300.Therefore, if this dilution you expect that it will give more than 300 colonies you should dilute thefluid further so that 1 ml will be expected to yield between 30 and 300 colonies.2-In the initial dilution (1:10) if no microbial colonies are recovered, express the result as "lessthan 10 m.o /ml of specimen".

1-Suspend 10.0 g(solid) or 10.0 ml(liquid) of the specimen influid soybean casein digest medium (in PH7.2 phosphate buffer) or  buffered sodium colored-peptone solution PH7.0to make 100 ml.It will be 1:10 dilution (1/10)
. It is calledpretreatment material
  N.B;  Add the specimen to medium not more than 1 hour after preparing the appropriatedilutions for inoculation.2-Prepare a series of 12 tubes each containing 9-10 ml of soybean-casein digest medium. 3-Divide these 12 tubes in four sets of three tubes each. 4-To each of the first three tubes add 1 ml of 1:10 dilution described in no 1. 5-To the next three tubes, add 1 ml of a 1:100 dilution described in no 4. 6-To the next three tubes, add 1 ml of 1:1000 dilution described in no 5. 7-To the last three tubes, add 1 ml of the diluent (as control( 8-Incubate the tubes at 30-35°C for at least 5 days  N.B; 1- No microbial growth should appear in the last three tubes. 2-If the reading is difficult, prepare a subculture in a liquid and evaluate the result after further period of incubation.9-Determine the most probable number of M.O per gm or ml of material using the next table.   N.B;  If, for the first column, the number of tubes showing microbial growth is two or less, themost probable number of M.O per gm or per ml is less than 100. References;
