1. INTENDED USE
The Rea T-Count reagent is a two color immunofluorescence reagentfor the labeling and identification of helper/inducer (CD3+CD4+) Tlymphocytes combined with a precise number of fluorescent countingbeads for absolute CD4+ and CD3+ T-Cell counts. This reagent isintended for flow cytometry based analysis in unlysed human wholeblood samples.
2. BACKGROUND
The Rea T-Count reagent contains fluorescently labeled antibodies thatbind to CD3 and CD4 antigens found on the surface of circulatingleukocytes. The CD3 antigen is a complex of at least six proteins knowncollectively as the T-cell receptor (TCR) complex. The antibody used inthis reagent binds to the 20kDa

chain of this complex. The CD4

antigen is a 59kDa protein. It interacts with class II molecules of themajor histocompatibility complex and is the primary receptor for theHuman Immunodeficiency Virus (HIV).Cells that are both CD3+ and CD4+ are identified as helper/inducerlymphocytes. Decreased CD4+CD3+ cell counts have been associatedwith some forms of immunodeficiency.

Rea T-Count Reagent
The Rea T-Count reagent contains dried down reagents formulated inbuffered saline, sodium azide and stabilizers. The reagent has CD4monoclonal antibody (clone RPA-T4) labeled with Phycoerythrin (PE)and CD3 (clone UCHT1) monoclonal antibody labeled with PE-Dyomics649.The monoclonal antibodies used in the Rea T-Count were assignedthese specificities at the 8th International Workshop on HumanLeukocyte Differentiation Antigens. A precise number of fluorescentcounting beads are included in each tube to allow single-platformdetermination of absolute CD4+ and T-cell counts. The Rea T-Count
