Carmine Method (Determine of boron) Apparatus 1-Spectrophothometer (585nm) 2-Dry test tube, soda glass/plastic Reagent 1-HCL , 6M *Mix

270 ml [HCL] + 230 ml distilled water 2-Carmine in [H2SO4 ], 0.05% (v/w) *1.25g carmine into freshly opened bottle of 2.5 L [H2SO4 ]. Shake for 2 days on shaking machine. 3-Boron standard solution *0.2860g boric acid into 500ml volumetric flask. Topup with distilled water to the mark.(100mg/l) *prepare 1mg/l and 5 mg/l 4-[H2SO4 ] Procedure 1-weight 1g sample and dissolve with distilled water in 1l flask. (500 ml first shake well and mark up to 1l) 2-pipette 10 ml of solution into 500 ml flask and mark up to volume. Pipette 2 ml of diluted sampleinto a dry test tube. 3- Add slowly 10 ml [H2SO4 ].Cover the test tube and let it cool. 4-when cool add 10 ml of Carmine in [H2SO4 ], 0.05% (v/w).Mix it well-pour one test tube to anothermcm teh tarik. 5- Let it 2 hour (colour development) (yellowish) 6-Measure absorption at 585 by using spectrophotometer. 7-Use a standard to calibrate spectrophotometer. Calculation Total boron B2O3 = [Boron]in mg/l x Dillution Factor x 0.322 weight (g) of sample taken
