A. PURPOSE : To check the existence oI Albumin and Glucose Levels in the urine.
B. TOOLS AND MATERIALS 1. Urine 2. Biuret
3. Fehling A and B (Benedict) 4. Measuring Cup
5. Test Tube 6 . Pipette
. Bunsen 8. Clamp
9. Matches
C. METHODS 1. Albumin Test O Prepare the materials and tools O Put urine slowly into measuring cup until it reaches the height oI 2 cm. Use a pipette to take an urine drops.
O Used as drops oI Biuret 5 drops
O Shake and observe color change. II the urine contained albumin, the color oI urine will turn purple
BeIore AIter 2. Glucose Test Enter the urine into a test tube, as high as 2 cm. Use the drop pipe to take the urine
Used as drops oI Fehling solution A and B each 5 drops. Shake and mix until evenly
Heat up the end oI the test tube over the Bunsen Ilame. heating process must be intermittently liIted so as not to burn. Do not let boil. Heating time 3-5 minutes
BeIore Burning AIter Burning
Observe the color change. II the urine contains glucose, urine color will be red brick
BeIore AIter
URINE TEST RESULTS TABLE Reagent Urine Original color Final Color Biuret

Clear Yellow
Bluish Green
QUESTIONS : 1. Please list the characteristics of healthy urine! O It has yellow clear color O It doesn`t contain oI glucose or albumine O It`s not muddy O It`s not contain oI blood, because iI it contains blood it means that something gets inIection in kandung kemih . How to means whether if the urine contained albumin? Albuminuria . istinguish the color of urine before and after the mixed reagent! a. Urine mixed with biuret reagent : - BeIore : Clear Yellow - AIter : Clear Yellowish (depend on the conditions oI urine)
b. Urine that have been mixed by reagent benedict (Fehling A and B) - BeIore : Clear Yellow - AIter : Bluish Green (depend on the conditions oI urine)
. What happens with urine if it`s positive diabetes? AIter the drops with Fehling solution A and B, and burned, the urine is yellow brown
. State the factors that influence the amount of urine! O The amount oI water drunk O Renal nerve stimulation that causes narrowing oI blood vessels leading to glomerulus O Antidioritic hormone (ADH) O Concentration oI insuline hormone O Levels oI salt that must be removed Irom the blood to keep the osmotic pressure O A person suIIering Irom diabetes mellitus (diabetes) glucose expenditure Iollowed by the increase in urine volume
CONCLUSION 1. II urine is mixed with Biuret and changed into purple or ther`s ring oI purple in surIace oI mixture, it means that mixture is positive to contain oI albumine or protein. II that happens, the person positive get Albuminuria. It`s negative to contain protein iI the color is clear yellow 2. II urine is mixed with Benedict and changed into red brick or Iirst color is yellow orange, it means that mixture is positive to contain oI glucose. II that happens, the person get Diabetics. It`s negative to contain protein iI the color is clear blue 3. Urine is inIluenced oI some Iactors O The amount oI water drunk O Renal nerve stimulation that causes narrowing oI blood vessels leading to glomerulus O Antidioritic hormone (ADH) O Concentration oI insuline hormone O Levels oI salt that must be removed Irom the blood to keep the osmotic pressure O A person suIIering Irom diabetes mellitus (diabetes) glucose expenditure Iollowed by the increase in urine volume
