Preliminary methods:
Double pithing
Exposure of the Heart
Setting-up of the Kymograph

A. Effect of Temperature
After exposing the heart, it was dropped with room temperature
ringer’s solution, cold water from the water dispenser, and hot water from the
water dispenser. With an interval of 2 minutes each and the ringer’s solution
being the baseline. Data obtained from each were recorded in the
kymograph.
B. Effect of Tension
Adjusting the clamp upward, Tension was increased by stretching the
muscles of the heart and the data was recorded in the kymograph
C. Effect of Drugs
a. Acetylcholine
2-3 drops were applied directly to the heart and the data was
recorded in the kymograph
b. Pilocarpine
2-3 drops were applied directly to the heart and the data was
recorded in the kymograph
c. Atropine + Acetylcholine

All the data and changes were recorded in the kymograph. 2-3 drops of Acetylcholine were applied directly to the heart. After 30 seconds. All the data obtained from the kymograph were compared and analyzed.2-3 drops of Atropine were applied directly to the heart. .
