Human Adipose Mesenchymal Stem Cells (MSCs)

HumanKine® Activin A
(Cat. No. GF400) Human cell-derived recombinant growth factors, such as Activin A, are preferred to bacterial cell-derived proteins for stem cell research, because they exhibit conserved posttranslational modifications and exert higher biological activities. HumanKine® Activin A, cited in a study of embryonic stem cell (ESC) miRNAs1. By measuring miRNA levels in nine different ESC lines, the authors found that miRNAs are promising markers for defining potency status with high resolution.

Description AhR Antagonist II, SR1

Details A cell-permeable purine compound that acts as a high affinity AhR antagonist (IC50 = 127 nM against 3 nM TCDD-induced luciferase reporter activity in HepG2 cultures), but not a panel of 61 kinases. SR1 treatment results in enhanced and sustained CD34+ population in long-term proliferating/expanding cultures of cord blood-derived HSCs (hematopoietic setem cells) in serum-free media containing Tpo, SCF, Flt3 Ligand, and IL-6 in a reversible manner. A cell-permeable pyrrolopyridinyl-benzamide compound that is reported to enhance the efficiency of 4F/4TF- (OSKM; Oct44, Sox2, Klf4, c-Myc) driven iPSC generation from MEFs cultured in either feeder cell-containing ESC media or in feeder-/serumfree iSF1 media. A cell-permeable imidazopyridinamine compound that selectively and potently induces differentiation of mesenchymal stem cells (MSCs) into hepatocyte-like cells (~47% at 5 µM). Within 14 days of treatment with SJA710-6, the differentiated cells exhibit typical morphological characteristics of hepatocytes and are able to store glycogen, secret albumin, produce urea, and uptake LDL. A phenothiazine class antipsychotic and dopamine receptor (DR) antagonist that is reported to exhibit CSCs- (cancer stem cells) differentiating (EC50 ≤9.4 µM for v1H9) and anti-leukemic activity, without affecting non-neoplastic H9 hESC, adult fibroblast-derived iPSC, or HSPCs (hematopoietic stem-progenitor cells). A cell-permeable acylurea compound that acts as an antagonistic Smo ligand (IC50 = 120 nM against 5 nM Bodipy-cyclopamine binding to murine Smo-transfected HEK293 cells). A cell-permeable hexahydropyrimidine compound that acts as downstream Hedgehog (Hh) pathway-selective blockers and target Gli-mediated gene transactivation (IC50 ~5 µM) in SAG-stimulated Shh-L2 cells, and exhibits in vivo antitumor efficacy. Description CBFbeta Activator, Kartogenin Pyrintegrin Details A cell-permeable biphenylcarbamoylbenzoate compound that potently induces the differentiation of human mesenchymal stem cells (hMSCs) into chondrocytes (EC50 = 100 nM). A cell-permeable pyrimidine compound that promotes the adhesion of individually dissociated hESCs on matrigel- or laminin-, but not gelatin-coated surfaces and substantially reduces trypsinization-induced apoptosis, while maintaining cells’ self-renewal ability and the pluripotency of expanded cultures for more than 40 passages. A cell-permeable indole compound that, when added (10 µM) to murine D3ES ESC (embryonic stem cell) cultures in the absence of serum and MEF (murine embryonic fibroblast) feeder cells, serves as a cheaper, chemical alternative to LIF (1,000 units/mL) for expanding ESCs in their undifferentiated state. A cell-permeable, orally available triazole-class antifungal agent that, in addition to its known inhibitory activity against fungal cytochrome P450 14-α-demethylase, also acts as an effective Hh signaling inhibitor (IC50 ~ 0.8 µM against SHHNinduced Gli transcription activity in Shh-Light2 cells) in a Ptch-independent manner. Description Aurora Kinase Inhibitor XI, HOI-07

Details A cell-permeable oxindole compound that selectively inhibits Aurora B, but not Aurora A, activity (IC50 ≤100 nM; [ATP] = 100 µM) and potently suppresses cellular histone H3 Ser10 phosphorylation (IC50 <100 nM in A549 cells; 24 h). Potently inhibits anchorage-independent growths of human lung and colon cancer lines (by >80% at 0.5 µM) in vitro. Purity: ≥97% by HPLC. A cell-permeable indandione compound that inhibits the Bmi1-Ring1A E3 complex ubiquitin ligase activity both in cell-free assays (by 100% against self- and Top2a ubiquitination at 25 and 50 µM, respectively) and in HeLa cells (by 100% against Bmi1 ubiquitination at 50 µM). Purity: ≥99% by HPLC. A cell-permeable benzodiazepine that suppresses CBFa/Runx1-CBFb transactivation activity (25 µM; 293-0 cells) via direct bindings to both subunits of the heterodimeric transcription factor complex. Shown to inhibit oncogenic fusion CBFB-MYH11-, TEL-RUNX1-, and RUNX1-ETO-, dependent leukemia cells proliferation (IC50 = 1.1 to 21.7 µM) in vitro. Purity: ≥98% by HPLC. (Cat. No. 480073) Programmed necrosis, autophagy and apoptosis are the three interconnected pathways that lead to programmed cell death. EMD Millipore recently made available Necrosulfonamide, a small molecule that irreversibly inhibits programmed necrosis by binding to the human mixed lineage kinase domain-like (MLKL) protein, a newly discovered member of the programmed necrosis pathway. Recently, Sun and colleagues identified Cys86 of MLKL as the Necrosulfonamide-binding site and showed that it blocked necrosis downstream of RIP3 activation2. Rho Inhibitor, Rhosin
(Cat. No. 555460) Rhosin, a new cell-permeable compound that directly targets Rho GEF binding domain (Kd = 354 nM) and prevents Rho from interacting with its GEFs, is now available from EMD Millipore. Rhosin blocks the endogenous RhoA activity and growth of MCF7 breast cancer cells without affecting the non-tumorigenic epithelial MCF10A cells. Recently, Shang and colleagues showed that Rhosin specifically inhibited RhoA activity and RhoA-mediated cellular function without affecting Cdc42 or Rac1 signaling activities3. By suppressing RhoA activity, they could inhibit mammary sphere formation by breast cancer cells (EC50= 30-50 μM) and suppress invasion of mammary epithelial cells.

Details A dimethylamino substituted succinamate that, in addition to its well known use as a plant growth regulator, also acts as an inhibitor against histone lysine demethylases KDM2A, LSD1 (KDM7A; KIAA1718), and PHF8 (IC50 = 1.5, 2.1, and 0.55 µM, respectively). Purity: ≥98% by NMR. The H3K27me3 demethylase inhibitor GSK-J1 (Cat. No. 420204) against JMJD3 and UTX is made cell-permeable by ethyl ester derivatization to facilitate cellular delivery ([medium GSK-J4]t=0 = 30 µM; [cellular GSK-J1] t=1h = 1.6 µM). Simultaneous inhibition of cellular JMJD3 and UTX by GSK-J4 treatment is shown to block LPS-induced production of several proinflammatory cytokines in primary human macrophage cultures. Purity: ≥98% by HPLC. A benzothiadiazolyl-cyclohexenylethylurea compound that binds to protein arginine methyltransferase 3 (PRMT3) at the dimer interface and selectively inhibits its activity in an allosteric manner (IC50 = 1.6 µM). Although this compound is cell-permeable, it is metabolically unstable and therefore not recommended for cell cultures or in vivo applications. Purity: ≥99% by HPLC.

(Cat. No. ABS310) EMD Millipore’s new polyclonal antibody against human Carboxylesterase 1 is featured in a 2011 study of neurotoxins4. The major function of hCE-1 is to metabolize a variety of distinct harmful endogenous and exogenous compounds. Due to its similarity in structure and catalytic action with acetylcholinesterase, it is an attractive target for studying the effects of nerve agents and environmental toxins. Recently, Hemmert and colleagues created a highly active mutant form of hCE-1 by V146H and L363E substitutions and used this antibody to determine the specific activity of normal and mutant enzymes. (Cat. No. MABS330) A recently published proteomics study cited EMD Millipore’s new protein G purified monoclonal antibody against Secreted Frizzled-related Protein 2 (SFRP2). SFRP2 plays a significant role in embryonic development and in several pathological conditions, including myocardial disorders and cancer. Due to its sequence homology with the Wnt-binding domain of Frizzled, it acts as an antagonist of canonical Wnt signaling. In the paper titled: “Label-free LC-MSMS analysis of vitreous from autoimmune uveitis reveals a significant decrease in secreted Wnt signalling inhibitors DKK3 and SFRP2”5 Hauck and colleagues reported the use of anti-SFRP2 to show a very significant decline in SFRP2 levels in the vitreous humor of the eyes of horses suffering from the blinding disease, equine recurrent uveitis (ERU). This indicated that Wnt signaling was severly impaired in this disease. They also showed that although SFRP2 is stably expressed in ERU retina, it is lost at the outer limiting membrane.

Orally active in mice and exhibits NEFAc cholesterollowering efficacy in overnight fasted C57B16/J mice (ED50 ~100 mg/kg, p.o.) in vivo. Purity: ≥98% by HPLC. A benzothiazole compound that acts as an ATP competitive Dyrk inhibitor (IC50 = 0.24 µM and 0.23 µM for Dyrk1A and Dyrk1B, respectively), and displays >90% inhibition on DYRK2, DYRK3, CLK1, CLK4, casein kinase 1 (CSNK1D), and PIM1 among a panel of 66 kinases, in an in vitro screening assay. Purity: ≥98% by HPLC. Purity: ≥95% by HPLC. Methysticin has been identified from kava to have potent NF-kB inhibitory activity and minimum toxicity. No. 537000) Dual-specificity tyrosine phosphorylationregulated kinase 1A (Dyrk1A) is a regulator of neuronal development, and therefore, small molecule modulators of this enzyme are useful for studies of neuroregeneration. EMD Millipore has now made available proINDY, a selective inhibitor of Dyrk1A, ProINDY was used by Ogawa and colleagues to inhibit overexpressed Dyrk1A in Xenopus embryos, thereby rescuing developmental defects6. Their results indicate that chemical genetic analyses may shed light on cell fate pathways affecting neurodegenerative diseases and congenital brain defects.
