12. Research interests, possible project ideas and what you hope to gain from this
experience.
Research interests:
Title: 8-OHdG – a novel Biomarker to evaluate Oxidative DNA damage in Micro
vascular Complications.
Description: Vascular complications are the leading cause of morbidity and mortality in
patients with diabetes. In nuclear and mitochondrial DNA, 8-hydroxydeoxyguanosine (8-
OHdG), an oxidized nucleoside of DNA, is the most frequently detected and studied
DNA lesion. Upon DNA repair, 8-OHdG is excreted in the urine. Numerous studies have
shown that oxidative DNA damage links pathogenically to a variety of aging-associated
degenerative diseases such as cancer, coronary heart disease and diabetes. Quantification
of urinary 8-OHdG, a specific DNA repair products in the urine, can be made with a
simple ELISA. Finally, the study of Urinary 8-OHdG can lead to predict the earlier onset
of vascular complications.

Project ideas:
Title: Synthesis and Characterization of Cerium oxide Nanoparticle to study the
hyperglycemia induced Endothelial Dysfunction in Cultured Bovine Retinal Endothelial
Cells.
Description: In order to combat the hyperglycemia induced Endothelial Dysfunction in
humans, I have proposed that rare-earth cerium oxide (CeO2) nanoparticles (free-radical
scavengers) protect normal tissue from hyperglycemia induced damage. Earlier studies
and reports suggest that these nanoparticles may be a therapeutic regenerative
nanomedicine that will scavenge reactive oxygen species, which are responsible for such
induced cell damage. In the end, CeO2 nanoparticles provide a tremendous potential for
endothelial cell protection and can lead to significant benefits for the preservation of
human health and the quality of life for humans having Diabetes Mellitus.

Gain from this experience:


1. Gain better understanding of the scientific process and the application of the
knowledge gained from basic science research and state of the art techniques to solve
complex problems in biology and medicine.
2. Like to be a member of a research community working at the forefront of science,
medicine and biotechnology.

work, techniques exposed to and past lab experience.
Research experience:
Completed a project entitled “Induction of Apoptosis by Elevated levels of
Glucose through Caspase -3 Activation in Bovine Retinal Endothelial Cells” for a period
of four months at Animal Cell Culture Laboratory, Kalasalingam University, Tamil
Nadu, India.

Technical background:
Familiarized with the theoretical concepts and the practical applications of various
subjects like Bio-Nanotechnology, Nanolithography, Molecular Biology, Microbiology,
Animal Biotechnology, Plant Biotechnology and Immunology.

Internships:
Completed a project entitled “Strain improvement of Streptomyces.sp to
increase production of small molecules- Immunomycin” at Research and
Development Department, BIOCON LIMITED, Bangalore, India, from May 15th 2008
to August 16th 2008.

important cells and their Molecular Characterization” organized by PSG
College of Technology Coimbatore and Supported by Department of
Biotechnology(Ministry of Science and Technology) from November 9th to

Department, PHARMAFABRIKON Madurai, Tamil Nadu, India, from June
21st to June 27th 2006.
3. International Seminar and Workshop on Medical and Pharmaceutical
Nanotechnology held at Anna University Tiruchirappalli, Tamil Nadu, India
from September 25th to September 26th 2008.
4. National Workshop on MEMS in Biomedical Applications held at SSN College
of Engineering Chennai, Tamil Nadu, India from September 17th to September
18th 2009 .

Techniques exposed to and past lab experience:


1. High Performance Liquid Chromatography
Measured the activites of various Anti-Cancer molecules such as Tacrolimus,
Orlistat and Hygromycin through HPLC.

2. Cell Cytotoxocity Assays


The cells harvested from animal tissues and organs are analyzed for measuring
apoptosis. (Caspase and MTT Assay)

3. Maintenance of Industrially Important Cells and their Molecular


Characterization of Cells
Cryopreservation, Isolation of DNA, PCR-RFLP, RAPD, Multiplex PCR,
Western Blotting and PCR-Primer designing Bioinformatics tools.

4. Biochemical estimation on Type-II Diabetic patients


Biochemical estimation of Blood (Glucose, Creatinine, Urea and Hydrogen
Peroxide) and Urine (Glucose, Urea, Hydrogen Peroxide and Ketone bodies).
