The differentiation of primary human bronchial epithelial cells
(HBECs) to a physiologically relevant phenotype can only be
achieved using air-liquid interface (ALI) culture methods (Figure 1),
coupled with appropriately supplemented media. However, current
ALI culture methods often result in unsatisfactory and inconsistent
pseudostratification and mucociliary differentiation.
PneumaCult™-ALI is a serum- and bovine pituitary extract
(BPE)-free medium that has been developed to reproducibly
support improved mucociliary differentiation of primary HBECs
in ALI cultures. PneumaCult™-ALI is supplied with a detailed
protocol, enabling easy preparation of a fully differentiated
mucociliary epithelium from undifferentiated HBECs. The resulting
differentiated cultures exhibit physiologically relevant proportions
of goblet and ciliated cells, with widespread ciliary movement 21
to 28 days after air-lift (Figure 2). Cultures also feature improved
homogeneity of the thickness of the differentiated epithelial layer
when compared to traditional protocols.
