Introduction

The approval of Chinese hamster ovary (CHO)-derived tissue plasminogen activato


in 1986 revolutionized medicine and raised the possibility of using mammalian cell
culture for the manufacturing of protein therapeutic products. The reason why CHO
cells remained as the preferred mammalian cell line for the production of
recombinant protein therapeutic are (lai et.al., 2013):

HO cells are capable of adapting and growing in suspension culture which is

ideal for large scale culture in the industry


CHO cells pose less risk as few human viruses are able to propagate in them
CHO cells can grow in serum-free and chemically defined media which

ensures reproducibility between different batches of cell culture


CHO cells allow post translational modifications to recombinant proteins
which are compatible and bioactive in humans

ORIGINATOR

Pharmaceutical Cell Line Development was develop by ProBioGen to fulfill clients


needs. ProBioGen is a technology provider with focus on cell line engineering,
process development and Good Manufacturing Practice (GMP). CHO-DG44 and CHOK1 was use by ProBioGen as starter cell lines and both have been selected for most
efficient growth in chemically defined media and for optimized biopharmaceutical
manufacturing. It also immense adaptive ability and safer than actual human cell
lines because most pathogenic virus includes HIV do not infect these cells.

Chinese Hamster Ovary (CHO) cells are the most common cells applied in
commercial production of biopharmaceuticals. The original CHO cell line isolated in
the 1950s gave rise to a range of genetically different progeny, such as K1-, DukX
B11-, DG44- cell lines and others. The choice of CHO cell line can affect target
protein quality and achievable yield.
