1. We could measure the activity of the lactase enzyme by incubating our enzyme preparations
with the sugar lactose. Why is it important and useful to use ONPG instead?
ONP has an easy to see color change, and so it can give a visual indicator to show reaction
process, similar to a titration. The color is also useful in that spectrophotometry can be used to
gauge the light wavelength absorption due to the color, which allows us to quantify the reaction
progress.

2. In this experiment, we’ll incubate our samples at 37 C. Is this the temperature at which you’d
expect lactase to have the greatest activity? Why or why not?

2) Enzymes exist in our bodies, which generally have a temperature of 37 C. In order to make
the enzymes work as they would normally, the temperature should match that of their typical
habitat.
