What is Mass Spectrometry?
Mass spectrometry (MS) is an analytical technique that ionizes samples (solid, liquid, or gas)
and sorts the ions based on their mass-to-charge ratio. It is used to measure the characteristics
of individual molecules, by accelerating them and subjecting them to an electric or magnetic
field. After sorting, the resulting mass spectrum is a plot of the (relative) abundance of the
produced ions as a function of the m/z ratio. Because prior detailed knowledge of the sample is
not required, this technique is applicable to identify unknowns.
MS is widely applicable to environmental analysis, food contaminant and quality analysis,
isotope dating and tracking, proteomics, lipidomics, bioanalysis, definitive biotransformation,
metabolomics to clinical application and forensics/toxicology.
How does LC/Triple Quadrupole mass spec work?
A Triple Quadrupole Liquid Chromatography/Mass Spectrometry (Triple Quadruple LC/MS) begins with a liquid
chromatograph, which separates complex chemical mixtures into their pure constituents based
on the chemical properties of each compound in the mixture. The separated, pure compounds
then sequentially enter a mass spectrometer, which consists of two stages of mass analysis.
1. The ion source ionizes the sample and the ion products are mass analysed by the first
quadrupole assembly. However, in the case of a Triple Quadrupole LC/MS, mass-
selected ions exiting the first quadrupole assembly collide with an inert gas in the
second assembly (which may be of distinct geometries), and fragment to produce a set
of ions known as product ions. A chamber called the collision cell surrounds this
assembly and is typically pressurized with an inert gas.
2. The product ions undergo further mass analysis in the second quadrupole assembly to
detect selected ions. Two stages of mass analysis yield far greater chemical specificity
than a single stage, because of the system’s ability to select and determine two
discrete, but directly related sets of masses.
