4.1 Crude extract Five species of soft coral were taken from intertidal area of Siladen Island: Umbellulifera sp, Sarcophyton sp, Xenia sp, Lemnalia sp and

Capnella. Each soft coral was cut and soaked in methanol for 3 days. After, it was evaporated to remove methanol and the crude extract obtained was weighed. Table 02 describes precentage of crude extract obtained by methanol extraction. Table 2. Wet weight, Biomass and Crude Extract Species

Figure 13. Capnella sp (under water and open water) 4.2 Antibacterial activity The antibacterial test results show the difference in the diameter formation of clear zone by each crude extract against bacterial used. The antibacterial activity was higher for Umbellulifera compared to other soft corals tested. In general, the effective crude extracts inhibited the growth of all bacterial used, however the higher activity was obtained against Pseudomonas sp. (Table 03). Table 03. The test antibacterial results show that there were difference in the diameter formation of clear zone formed by each fraction (Umbellulifera sp. and Lemnalia sp) against bacterial used (Salmonella, E. coli, and S. aureus), The higher antibacterial activity was obtained on butanol fraction of Umbellulifera sp and in Ethyl acetate fraction of Lemnalia sp as sumerized in Table 4. The results show that clear zone formation by water dissolved faction of crude extract of Umbellulifera sp were varied between bacterial tested compared to clear zone formed by amphycillin. Salmonella sp 130%, E. coli 0%, and S. aureus 75%. While for Butanol dissolved faction S. aureus had a higher activity of 280%. Salmonella sp 180%, and E. coli 58%. In contrast with above results, when Ethyl acetate was used as a dissolving 39

agent the activity of antibacterial tend low against all bacterial tested. The antibacterial activity was obtained only in E. coli (52%), while form Salmonella and S. aureus were no activity obtained. The water dissolved crude extract of Lemnalia sp was obtained positive ainst E. coli (52%) while no activity obtained against Salmonella and S. aureus. The same results were obtained when butanol was used as a dissolving agent. In contrast with above results, ethyl acetate dissolved faction show that antibacterial activity was higher against E. coli (111%), followed by S. aureus (81%) and Salmonella (80%), respectively. Figure. 19 -24 show the formation of clear zone by crude extract against bacterial used.
