SEQUENCING METHOD

ABSTRACT

This study aims to determine the genetic diversity of the growth hormone gene in Kacang Goats using

the Sequencing method. This study used 96 samples of blood from Kacang Goats. DNA was isolated

from each blood sample and then continued with the use of the PCR method and the Sequencing

method. The diversity of growth hormone (GH) genes is seen from the results of Sequencing (SNP).

There are 8 polymorphisms in the Exon 1 Intron 1 GH gene in Kacang Goats, Six mutations were

detected, namely the G → T mutation at -257, the G → A mutation at -237, the G → T mutation at -231,

the A → T mutation at position -81, mutation C → T at position -31, and mutation G → A at position 20

with successive genotype frequencies 0.01; 0.01; 0.04; 0.08; 0.10; 0.7. Two insertions were also found,

namely insertion A detected at position 4 with the allel frequency of -298 and insertion T at position -298.

The results of this study indicate that the GH gene in Kacang Goats has a high diversity, so that further

research is needed to determine whether this diversity is related to the nature of production.
