Test Procedure 2. Culture bottles to hold 60 mL sample volume
1. Enrich food samples. 3. Modified peptone
2. Add 0.5 mL of enriched samples, negative and positive controls 4. Modified Giolitti-Cantoni broth
into separate 12 X 75 mm glass tubes. 5. Water bath at 37°C
3. Add 0.1 mL pretreatment reagent to each tube. Shake the tubes for 5 6. Water bath at 65°C
seconds, then incubate the tubes in 37°C water bath for 30 minutes. 7. Graduated cylinder
4. While in water bath add 0.1 mL lysis reagent to each test tube. 8. Absorbent paper
Remove rack of tubes and shake for 5 seconds. Replace in bath 9. Test tubes, sterile with caps to hold 3 mL sample volume
and incubate in 37°C water for 45 minutes. * See Equipment and Accessories, pages 91–95
5. Add 0.1 mL of S. aureus probe solution to each tube.
6. Place dipsticks into sample tubes. Raise and lower dipsticks 5 times
to mix and incubate at 65°C for 1 hour.
7. Wash dipsticks two times for 1 minute each, first at 65°C, then at
room temperature.
8. Transfer the dipsticks to tubes containing 0.75 mL of enzyme
conjugate and incubate at room temperature for 20 minutes.
9. Wash the dipsticks two times for 1 minute each at room temperature.
10. Transfer dipsticks to tubes containing 0.75 mL of substrate-chro-
mogen and incubate at room temperature for 20 minutes.
11. Remove the dipsticks and add 0.25 mL of stop solution.
