INTRODUCTION
• A number of human disorder can be
traced to the absence or malfunction of
a protein normally synthesized in the
body.
• Animal protein are there fore used
whenever these are not effective but
there are not many disorders that can
be treated with animal proteins.
• There is always the possibility of side
effect such as an allergic response.
• Recombinant DNA technology provides
a powerful tool for new pharmaceuticals
product development and production.

Human insulin the first FDA-approved rDNA drug in
1982 for the treatment of insulin-dependent diabetes.

 Insulin synthesized by the beta-cell of the islets of
Langerhans in the pancreas controls the level of
glucose in the blood.
 Insulin deficiency manifests itself as diabetes mellitus.
 Limited production of human insulin required the
hormone from the animal’s like cow and pigs.
 Although animal insulin is generally satisfactory but

• INTRODUCTION :-
In the human body the
malfunctioning of growth hormone
that is SOMATOSTATIN and
SOMATOTROPHIN leading to painful
and disabling disorders such as
Acromegaly(uncontrolled bone
growth), and Dwarfism.

SYNTHESIS OF GROWTH
HORMONE
 SOMATOSTATIN:- It was the first
human protein synthesized in E.coli .
 It is very short protein,only 14 amino
acid in length that’s why it was
suited for artificial gene synthesis.
 SOMATOTROPHIN:- This protein is
191 amino acid long.
 cDNA cloning was used to obtain a
somatotrophin-producing E.coli stain.

RECOMBINANT FACTOR VIII
• Factor VIII is responsible for Blood Clotting.
• The inability to synthesize factor
VIII,results in causing a disease called
Haemophilia.
• It leading to a break down in blood clotting
pathway.
• The factor VIII gene is very large,over 186
kb in length and split into 26 exons and 25
introns.
• mRNA codes for large polypeptide which
undergoes complex series of post
translational modification.
