1. E. coli was incubated with aeration in a nutrient medium containing two carbon sources, and the following growth curve was made from this culture.

log10 of no. of cells

a. Explain what happened at the time x.
At the time x, the E. coli has depleted the glucose carbon source, therefore shifting there meal to their least preferred food, the lactose . E. coli prefer to consume glucose and only use lactose after the glucose has been depleted.

b. Which substrate provided “better” growth conditions for the bacteria? How can you tell?
The glucose provided the better growth conditions for the bacteria than the lactose. In the graph, during the consumption of glucose by the E. coli there is a great increase in the number of cells. After the glucose is depleted, the E. coli utilizes the lactose as a carbon source there is a decrease in the number of cells than that of when they consumed the glucose because glucose is a

2. Clostridium and Streptococcus are both catalase-negative. Streptococcus grows by fermentation. Why is clostridium killed by oxygen, whereas streptococcus is not.
Clostridium is an obligate anaerobes which means they are unable to utilize oxygen for energy yielding reactions. They lack enzymes to neutralize harmful forms of oxygen that is why they cease to exist in the presence of oxygen, while streptococcus is an aerotolerant anaerobes that grows by means of fermentation(anaerobic growth). Oxygen has no effect because of the presence of an enzyme called superoxide dismutase(SOD) that allows harmful forms of oxygen to be partially neutralize.
