How to calculate Ic50


The IC50, the concentration of substance that provides 50% inhibition to certain reaction, is most
commonly used to represent the inhibitory effect of compounds on competition binding assays and
functional antagonist assays. In simpler terms, it measures how much of a particular substance/molecule
is needed to inhibit some biological process by 50%.
In similar terms, there is the EC50, which is used to describe the effect of compounds on
agonist/stimulator assays. The EC50 shows the concentration giving 50% of that compoundÅfs maximal
response.

Before fitting a dose-response curve to obtain the EC/IC50, each well should be converted to either
percent activity or percent inhibition with respect to positive and negative controls. Then all replicate
wells from a given run (including multiple plates per run) for a given concentration should be averaged
either by taking the mean, or preferably, taking the median.

To start the calculation of IC50/EC50 (on the web, it was described as IC50 for simplicity), input your
data in 2 (x and y) columns into the box. Alternatively, you can import your data from a file. The file
format should be tab-delimited text, which can be easily generated by using the software Microsoft
Excel. After importing your data, just click "calculate".

The four parameter logistic model (4PL), also called the Hill-Slope model, is used to calculate IC50 on

In results window, you can see your input data, file name, date of calculation, semi-logarithmic plot of
your data, and IC50 (important!). In addition, there is two positions indicated as "sequence". It is not
directly related to your results. However it is better to check whether the values of both "sequence" are
same, which means the calculation program had worked correctly.
