For the isolation and culture of zoosporic fungi using baiting techniques, baits were

obtained from dead insects like cockroaches. Aside from dead insects, shrimp exoskeleton ,
snake skin, and pollen grains were also acquired for this experiment. This array of baits
includes several types of keratin, some chitinous materials, and some cellulose or cellulose
derivatives. The group were able to obtain three baits representing each of keratin, chitin and
cellulose materials. Pond waters with soil or plant debris were also callected and placed inside
sterile, deep-based petri dishes. Baits were placed on the pond water-filled petri dishes and
incubated at room temperature under cool conditions. All baits were then examined under the
microscope after two days and daily after the first day of observation. The pollen grains were
examined after 12 hours after culture and frequently thereafter. Plates were either directly
viewed under the scanning objective of microscope for culture observation. Samples were
also readily obtained by lowering a dry, clean cover slip, faced down onto the water surface
and quickly withdrawn. The cover slip was then placed directly on glass slides with a drop of
water and observed under the microscope. A small portion of the baits were also placed on a
glass slide and was directly observed under the microscope. The hyphae, reproductive
structures, and zoospores were observed and these observations were recorded. For the
preservation of the materials, a drop of lactophenol mounting medium with acid fuchsin was
added to the edge of the cover slip and was allowed to replace the water. The permanent
mount was prepared and submitted to the laboratory instructors.
