determining the precise order of nucleotides
in a DNA molecule.
● It unveils the specific order of adenine (A),
cytosine (C), guanine (G), and thymine (T)
along the DNA strand.
● This sequence carries the genetic
information vital for the growth,
functioning, and reproduction of organisms.
● To decode the genetic blueprint, aiding in
understanding traits, diseases, and
evolutionary connections.
NANOPORE
Nanopore sequencing utilizes tiny nanopores, which are minuscule channels at the nanometer scale, to sequence
DNA molecules.

❏ Biological Nanopores- Transmembrane protein channels


(alpha hemolysin)
❏ Solid state Nanopores- Silicon Nitride or holed Silica plate
filled with Fe
The magnitude of current depends on base composition (and size,shape of pore)
Place the nanopore (artificial) in a conducting solution

Apply voltage

Add nucleotides

Drive the nucleotides through the nanopore (electrophoresis)

Characteristic pattern of change in the pattern of current

Nucleotide: A T G C
Magnitude: ←↘ ↗ →
Use electrophoresis or Motor proteins to pull the nucleic acid through the nanopore
