Antimicrobial Activity of Lumbricus rubellus Protein Against Escherihia

coli

ABSTRACT
Lumbricus rubellus worm has been used by people as the medicine. This worm is believed to
have activity against thypoid. This research was done to determine the antimicrobial activity of
protein of Lumbricus rubellus against Escherichia coli bacteria. The protein was obtained from
the isolation of dried worms are sold in the market. The method used in this research are 4
stages. First dried worm was extracted by using Nematodes Solubilization Buffer and
centrifuged to obtain supernatant. Second the supernatant was fractionated by gel filtration
chromatography. Third fraction was identified by HPLC detector and Liquid Chromatography
Mass Spectroscopy. Fourth test of the activity of the three fractions that have the highest
absorbance is 0.643, 0.527 and 0.434 with the disk diffusion method. The results showed that
there was no antimicrobial activity of the three fractions of the bacterium Escherichia coli. This
is evidenced by there were no inhibition zones on agar media.
