Summary There are 2 main objective of this experiment where the first one is to determine total viable aerobic

count in Labisa pumila and second is to determine the presence of E. coli in Labisa pumila. In handling this experiment there are two main part of this experiment, where the first part is preparation of sample and second part is testing. The experiment is start by weight in the sample and tenfold dilution is prepared. On second part there are two main sub test which is total viable cell and test for specified microorganism. For total viable cell there are two methods which is pour plate method and spread plate method. Both of this method will produce or show a lot colony if microorganism on its surface of the agar and the average number of colony is count. On the test for specified microorganism, the sample first incubates in nutrient agar in conical flask in 35C for 2 day, and then it is transfer to MacConkey agar and incubates in 43C for 2 day. Then it is subculture on the petri dish containing MacConkey agar and incubate in 35C for 3 days. The result is the growth of red, non- mucoid colonies of gramnegative rods indicates the possible presence of E. coli.

Conclusion Based on the experiment, it is prove that the objective of this experiment which is to determine total viable aerobic count in Labisa pumila and to determine the presence of E. coli in Labisa pumila is successful. The result of this experiment is shown a positive result which indicate that the sample is producing a standard result as in theory. However some of the testing need to further test, this is due to the most of the test that has been handle is only on the observation. If there is further test, each of the colony produce will be determine its type and its species. When it is determine, more specific function of microorganism can be known. Some of the procedure also can change in order to get more difference or in other word is one procedure to show a positive and another is show the negative result in order to get or differentiate the reaction happen.
