Light microscope, eye-piece graticule/ micrometer, stage micrometer Method The eye-piece graticule was placed into the ocular lens by removing the top lens and inserting it into the eyepiece. The stage micrometer, measuring in 1/100 mm was placed on the stage and arranged so light passes through the scale. The low power objective was clicked into position and focused onto the stage micrometer scale. The lines on the slide were aligned with those of the graticule. The ocular lens was rotated as necessary to facilitate the alignment. The number of lines on the stage micrometer along the length of the graticule were counted. The width of one eye-piece unit for the low power objective was calculated. The procedure was repeated for the other two objective lenses that needed to be calibrated. Results 1 eye-piece graticule unit = 0.34mm at magnification 40x 0.14mm at magnification 100x
