Antisera produced against an antigen contain polyclonal antibodies.

mABs that originate from a single clone, have a single specificity for Antigen.

The source of mAbs are somatic hybrids of myeloma cells and plasma B-cells isolated from mouse spleen.

mAbs of mouse origin, can cause hypersensitivity in humans. So, a new strategy is cloning human antibody constant regions to mouse Antibody-binding region; creating a hybrid antibody; highly specific but less likely to cause hypersensitivity reactions.

APPLICATIONS OF mAbS

1)TYPING OF TISSUES 2)IDENTIFICATION OF PATHOGENS 3)IDENTIFICATION OF TUMORS 4)IDENTIFICATION OF SURFACE ANTIGENS 5)CLASSIFICATION OF LEUKEMIA 6)IDENTIFICATION OF FUNCTIONAL POPULATIONS OF DIFFERENT TYPES OF T-CELLS 7)IDENTIFICATION AND MAPPING OF ANTIGENIC DETERMINANTS (EPITOPES) ON PROTEINS 8)IDENTIFICATION OF ANTIGENS; CONJUGATION OF mAbS TO COLORIMETRIC AND FLUOROMETRIC OR ENZYMATIC ACTIVITY 9)MICROBIAL TYPING
