    General Information:
    Terrific Broth was developed by Tartoff and Hobbs to increase yields in plasmid 1 bearing Escherichia coli . Terrific Broth is an enriched medium, which helps the recombinant strains of E. coli maintain an extended growth phase. The large amount of tryptone and yeast extract in the medium allows the bacteria to produce a higher plasmid yield. The tryptone and yeast extracts also provide additional nutrients and growth factors required for enhanced growth of recombinant strains of E. coli. Glycerol is added as an additional carbohydrate source in this medium. The potassium phosphates are present to prevent a drop in pH of the medium during bacterial growth. Formula: (Per Liter) Tryptone = 12.0g Yeast Extract = 24.0g Potassium Phosphate, Dibasic = 9.4g Potassium Phosphate, Monobasic = 2.2g Final pH 7.2 0.2 at 25C Storage: Dehydrated media should be stored in a dry area at 18-26C. Prepared media should be stored at 2-8C. Method of Preparation: 1. Suspend 47.6 grams of Terrific Broth powder in 1 L of purified water. 2. Add 4 mls of glycerol to the medium. 4. Autoclave at 121C for 15 minutes. 5. Allow the solution to cool to 50C or below before adding additional antibiotics or nutritional supplements..
