The paper you found looks like something you can work from. I wouldn't worry to much about how the set up is well thought out. That's great for you - less work you have to put in to creating a flow system. Your innovation is study how platelets are affected by surface nanotopography. You may also want to consider adding a protein layer on the surface of your patterns. Mary made a lot of comments since she has more experience with platelets, so I would look at those.
