Protocol for Replica Plating Adenine Mutants
Purpose:
To score whether or not the diploid unknowns can grow in the absence of adenine.
Hypothesis:
Cells will/will not grow based on their individual characteristics and the medium on
which they are plated.
Prediction:
If unknown strains grow/do not grow, then we will be able to determine their
mutant character. DV- growth of unknows, IV- unknown (mutant) yeast strains
Materials/Method:
1. Unknown (mutant) yeast strains
2. Plates of different media
3. Velvet for replica-plating
Protocol:
Label YPD plates (3) and SD-ade plates
Mate unknown strains onto YPD
Replica-plate onto SD-ade plates tomorrow

Effects of Ultraviolet (UV) Light on Yeast

Purpose:
To estimate the amount of UV exposure to yeast cells by measuring % survival after
subjecting yeast to UV exposure over different lengths of time
Hypothesis:
Over longer periods of time, % survival of yeast will decrease.
Prediction:
If yeast is subjected to UV exposure then % survival will vary inversely with length
of time exposed. IV- UV light, DV- % survival of yeast
Materials/Methods:
1. 12 YPD Plates
2. Tube of sterile H20 (15 ml)
3. Spreader plate yeast
4. Ethanol to sterilize spreader
5. Bunsen Burner
6. Brown paper bags, sharpie
7. 300 mL of 10 culture of 922 wildtype in a microtube
8. 150 mL of a 10 culture of rad 1 in a microtube
9. UV-B transilluminator box shielded with a large plexiglass
