Chlorophyll content by OD

During the experiment chlorophyll content was measured by spectrophometer


and flow cytometer. The measurement of flow cytometer is more accurate than
spectrophotometric measurement so in this part flow cytometric result was used
to evaluate the chlorophyll content.

Biomass concentration
Biomass concentration was analyzed by two different method which are
spectrophotometer and filtrating sample over glass fiber filters and measuring
the mass increase of the dried filters. Second method gives more reliable result
due to formation of biofilm and sediment during the fermentation.
During the nitrogen starvation, it is expected that cell concentration will remain
stable or increase litlle at the beginning of the experiment. Because of lack of
nitrogen cells are not able to produce protein and genetic materials for cell
division. In this part of the report only one photoperiod and only begining and
final biomass concentration was shown. More detailed information is in the
appendix.
In figure 1 it is easily seen that biomass concentration remained stable during the
fermentation. Similar trend was observed in other photoperiods. Only in low light
intensity shows diffrent trend. The possible reasons for that are improper mixing
and biofilm formation. Similar trend was observed in other photoperiods
