Background: Leukemia is one type of hematological malignancy marked with
uncontrolled proliferation. Cell line of HL-60 and K-562 is a progenitor cell
culture that can be used to detect leukemia. Nutgrass root is one of the potential
medicinal plants containing quercetin which can inhibit proliferation of cancer
cell.
Objective: The study aims at examining the effect of nutgrass root (Cyperus
rotundus. L) extract to inhibit proliferation of the leukemic cell line of HL-60 and
K-562.
Method: The study uses a posttest only control group design, with four (4)
experimental groups. Group 1 acts as the control group, Group 2 is treated using
the extract f nutgrass root at the dosage of 20ug/ml, Group 3 is treated using the
extract f nutgrass root at the dosage of 40ug/ml, and Group 4 is treated using the
extract f nutgrass root at the dosage of 80ug/ml; each group is observed after 48
and 96 hours of treatment at flowcytometri. The study is carried out for a month.
Data is analyzed using One Way ANOVA, Tukeys test 5%, correlation test, and
linear regression analysis.
Findings and Discussion: The results of oneway ANOVA sig=0.000 (p<0.05)
show some effects on the treatment using nutgrass root (Cyperus rotundus. L)
extract, in which it can inhibit proliferation of the leukemic cell line of HL-60 and
K-562. The results of the correlation test sig=0.000 (p<0.05) and regression
analysis sig=0.000 (p<0.05) show the significant correlation between the dosage
of the nutgrass root extract given with the number of proliferation of the leukemic
cell line of HL-60 and K-562.
Conclusion: The treatment using nutgrass root (Cyperus rotundus. L) extract has
been proven to effectively inhibit proliferation of the leukemic cell line of HL-60
and K-562.
