This is to certify that Muhammad Nasir has successfully completed a six month internship at my
laboratory in the Department of Dermatology at Yale University from July to December, 2014.
Muhammad came to our group to study a cohort of subjects he had collected in Pakistan, mobilizing
clinical resources to assemble an interesting group of subjects. In fact, he had already done considerable
‘work on these patients prior to coming to the lab, performing linkage analysis and pre-screening for
mutations, While here, he used clinical phenotypes to inform further screening of candidate genes,
identifying causative mutations in ~80% of patients. In four, he performed whole exome sequencing
analysis, finding a fascinating mutation in RSPOI which causes sex reversal and skin disease. His
analysis continues in three other kindreds and holds the potential to identify new genetic causes of skin
disorders.
In separate work, Muhammad has expressed interest in learning biological techniques utilized in
functional characterization of identified mutations. He has become proficient in primary culture of
keratinocytes and fibroblasts, the generation of skin equivalents, and retroviral delivery of tetracyeline-
regulated genes into primary keratinocytes. He has also become proficient in targeted mutagenesis,
immunostaining, Western blotting, and skin grafting in mouse models. He has done important work on a
novel, severe variant of Keratitis Ichthyosis Deafness (KID) syndrome which I expect will also lead to
publication.
During his time here, Muhammad participated in weekly laboratory meetings and departmental seminars
as well as in visiting professor lectures at the University. He presented his work effectively and was a
valuable contributor to our conversations.

hope that we will be able to continue to collaborate on this fascinating group of disorders. | fully expect
that he will have a productive career with adequate institutional support.
