ABSTRACT

Memory is an mental ability to recall back sensations and ideas. Spatial memory
has important role in foraging behavior. Dementia is images of several kinds of
diseases and conditions that grow when the nerve cells in the brain die or they
do not work functionally which are caused by oxidative stress. Antioxidant, as
electron compound supplier, is able to avoid any negative impacts of oxidant in
body by decreasing oxidative stress. Kurkumin has been proved that it has
advantages as antioxidant and anti-inflammation which can avoid dementia. The
purpose of this research is to know the kurkumin effect and the doses toward
spatial memory of mice dementia modeling which is inducted with TMT.
This research uses post test controlled design planning research using 30 mice
consist of 6 groups of mice, each group consists of 5 mice. In the first day until
the seventh day the group I is given kurkumin suspension with doses of 25
mg/KgBB, the group II is given kurkumin suspension with doses of 50 mg/KgBB,
the group III is given kurkumin suspension with doses of 100 mg/KgBB, the group
IV is given citikolin with doses of 200 mg/KgBB, the group V is given kurkumin
suspension CMC-Na 0.5% (sick control), the group VI is given suspension CMC-Na
0.5% (health control). In the eighth day, the whole groups are given TMT as
much as 8 mg/KgBB with I.P except the group I. The test data of spatial memory
obtained through Morris Water Maze test consists of acquisition trial, probe trial,
and censor motoric ability test. Then, they are analyzed statistically using
Kolmogorov-Smirnov test and continued using repeated measure with trust
degree of 95%.
The Morris Water Maze test result on the acquisition trial shows that there is
significant difference between health control and sick control (p=<0,05) which
means that the TMT giving of 8 mg/kg BB can cause cognitive function
decreasing. In the comparison of sick control group with the groups of 25 mg/kg
BB, 50 mg/kg BB, and 100 mg/kg BB shows that kurkumin can increase the mice
memorizing ability.
According to the research, it is obtained that kurkumin with doses of 50 mg/kg
BB can increase spatial memory of the mice modeling dementia that are
inducted with TMT.
