The cloning and function analysis of Csrl gene in

cucumber

The leaf is the main photosynthetic organ in plants and its shape is closely
associated with its photosynthetic function. Appropriate leaf shape improves
photosynthetic efficiency, elevates plant biomass, and decreases plant
disease. Therefore, it is important to understand the molecular mechanism
that controls leaf morphogenesis. We has screened a mutant with rolled
leaves (named M1545) by cucumber EMS mutant library. Genetic analysis
using the F2 population from a cross between this mutant and cucumber line
9930 indicated the mutation was controlled by a single recessive
gene(named Csrl).
1. Cloning of Csrl candidate gene
In order to map the locus for this mutation, primary mapping with SSR (simple
sequence repeats) markers, the mutated locus can be located the one
chromosome of cucumber.
Another F2 population with 150-200 rolling leaf plants was constructed from a
cross between Csrl and its original wild-type line CCMC. We will use MutMap to
identify the unique genomic positions most probable to harbor mutation
causing rolled leaves. Based on the region of primary mapping, We screen the
possible candidate Csrl gene.
