predictor of CHD
Method
• Hypolipidemic evaluation in rabbits
Feeding methodolgy
• White rabbits weighing 1.5-2.0kg
• Divided in to 4 groups 7 of each
• Atherogenic diet is given – wheat flour+
milk powder ,dried egg yolk, hydrgenated
fat, butter, dried yeast, salt, sucrose and
vitamin mixture.
Experimental design
• Group I : (control) vehicle treated for 60 and 120 days
• Group II : atherogenic diet + cholesterol feeding
(400mg/kg per rabbit per day) for 60 and 120 days
• Group III : atherogenic diet + cholesterol feeding for 60
days ; atherogenic diet withdrawn: controled diet + test
drug from 61 to 120 days
• Group IV : atherogenic diet + cholesterol feeding + test
drug from 1 to 120 days
Serum collection
• After 60 days blood is drawn from the marginal ear vein
• On day of termination of experiment i.e. 121 day the
animal are sacrificed , blood is collected through cardiac
puncture and analysed for hematological parameters.
• Serum is separated and stored at -20ºc until assayed
• Tissues are cleared of adhering fat, weighed on a torsion
balance and half of them are frozen for biochemical
studies while other half are fixed in 10% formocalcium
fixative for histopathological studies.
Serum analysis
• Serum is analysed for :
Total cholesterol
Triglycerides
Phospholipid
HDL cholesterol
LDL cholesterol
VLDL cholesterol
HDL ratio
> Atherogenic index = LDL+ VLDL / HDL
Evaluation of results
• Serum biochemistry , tissue biochemistry and planimetric
data are collected
• Test data are compared with control and Standard drugs
