    Abstract
    TounderstandhowYidCandSecYEGfunctiontogetherinmembraneproteintopogenesis,
    insertionandfoldingofthelactosepermeaseofEscherichiacoli(LacY),a12transmembrane
    helixproteinLacYthatcatalyzessymportofagalactosideandanH(+),wasstudied.Although
    boththeSecYEGmachineryandsignalrecognitionparticlearerequiredforinsertionofLacYinto
    themembrane,YidCisnotrequiredfortranslocationofthesixperiplasmicloopsinLacY.Rather,
    YidCactsasachaperone,facilitatingLacYfolding.UponYidCdepletion,theconformationofLacY
    isperturbed,asjudgedbymonoclonalantibodybindingstudiesandbyinvivocrosslinking
    betweenintroducedCyspairs.DisulfidecrosslinkingalsodemonstratesthatYidCinteractswith
    multipletransmembranesegmentsofLacYduringmembranebiogenesis.Moreover,YidCis
    strictlyrequiredforinsertionofM13procoatproteinfusedintothemiddlecytoplasmicloopofLacY.
    Incontrast,theloopsprecedingandfollowingtheinsertedprocoatdomainaredependenton
    SecYEGforinsertion.Thesestudiesdemonstrateclosecooperationbetweenthetwocomplexes
    inmembranebiogenesisandthatYidCfunctionsprimarilyasafoldaseforLacY.
