1. NI DUNG THC TP
nh tnh h th trong ch phm H th hon; khon ng
hoa, tri mu trong ch phm Bo ph khang.

2. KHI QUT V PHNG PHP SC K LP MNG


2.1. Nguyn tc
Phng php sc k lp mng (SKLM) c dng nh tnh, th
tinh khit v i khi c dng thc hin bn nh lng hot
cht.
SKLM l k thut tch cc cht c tin hnh khi cho pha ng di
chuyn qua pha tnh trn t hn hp cc cht cn tch. Pha
tnh l cht hp ph c chn theo tng yu cu phn tch, c
tri thnh lp mng ng nht v c c nh trn cc phin knh
hoc phin kim loi. Pha ng l mt h dung mi n hoc a
thnh phn c trn vi nhau theo t l quy nh trong tng
chuyn lun. Trong qu trnh di chuyn qua lp hp ph, cc cu t
trong hn hp mu th di chuyn trn bn mng theo hng di
chuyn ca pha ng vi nhng tc khc nhau. Kt qu ta thu

3. THC NGHIM
3.1. nh tnh H th
i tng nghin cu: Thc phm chc nng H th hon (l
200 vin) ca C s sn xut thuc YHCT Bo Ngha vi thnh phn
h th ch 40 g, phc linh 1 g, ng quy 8 g, k t 8 g, th ty t
5 g, ph c ch 3 g, ngu tt 4 g, xuyn khung 3 g, bch thc 8 g,
thc a 3 g, mc hng 3 g, sinh khng 3 g.
Phng php sc k lp mng:
Bn mng: Silica gel G
Dung mi trin khai: Ethyl acetat methanol nc (100:17:13)
Dung dch th: Ly 0,25 g bt dc liu, them 20 ml ethanol 96%
(TT), un hi lu cch thy trong 30 pht, ngui, lc, bay hi
n cn kh. Cn them vo 10 ml nc v 1 ml dung dch acid HCl
10% (TT) un cch thy 30 pht, ngui, sau lc vi 20 ml
ether ethylic (TT) 2 ln, dch ether c bay hi cn khong 1 ml
dng lm dung dch th.
Dung dch i chiu: Ha tan emodin trong ethanol 96% (TT)
c dung dch c nng 0,1%. Nu khng c cht i chiu th
dng 0,25 g bt H th (mu chun), chit nh dung dch th.
Cch tin hnh: Chm ring bit ln bn mng 5 l dung dch th v
dung dch i chiu. Sau khi trin khai, kh bn mng trong
khng kh nhit phng, quan st cc vt di nh sng t ngoi
bc sng 365 nm v hin mu bng hi amoniac.
Trn sc k ca dung dch th phi c cc vt pht quang cng
gi tr Rf v mu sc vi vt t c trn sc k ca dung dch
i chiu nu dng dc liu chun chun b dung dch i chiu;

3.2. nh tnh khon ng hoa

mu 200 mg, t b dip 160 mg, tang bch b 160 mg, bch b 160
mg, khon ng hoa 100 mg, tri mu 100 mg, ng sm 100 mg,
hnh nhn 100 mg, thin mn 100 mg, bc h 80 mg, hong cm 50
mg, t uyn 50 mg, cam tho 50 mg.

Bn mng: Silica gel G
Dung mi khai trin: Cyclohexan ethyl acetat (9.5 : 0.5).
Dung dch th: Ly 2 g bt dc liu, thm 2 ml ether du ha (40 oC
60 oC) (TT), ngm v thnh thng lc trong 1h. Lc, bc hi t nhin
n cn. Ha cn trong 1 ml cloroform (TT).
Dung dch i chiu: Ly 2 g bt Khon ng hoa ( muc chun),
chit nh mu th.
Cch tin hnh: Chm ring bit ln bn mng 20 l mi dung dch
th v dung dch i chiu. Sau khi khai trin, bn mng c
kh ngoi khng kh hay sy nh cho bay ht dung mi. Soi di nh
sng t ngoi bc sng 365 nm sau phun dung dch Vanillin
Acid sulfuric (TT), sy 105 oC cho n khi xut hin vt.Trn sc k
ca dung dch th phi c cc vt c cng mu sc v gi tr Rf vi
cc vt ca dung dch i chiu.
3.3. nh tnh Tri mu

mg, khon ng hoa 100 mg, tri mu 100 mg, ng sm 100 mg,
hnh nhn 100 mg, thin mn 100 mg, bc h 80 mg, hong cm 50
mg, t uyn 50 mg, cam tho 50 mg.

Dung mi khai trin: Cloroform ethyl acetate (11:1)


Dung dch th: Ly 5 g bt dc liu, thm 50 ml ethanol 95 % (TT),
un hi lu 60 pht, ngui, lc, ly dch lc, thm 2 ml dung dch
acid sulfuric 5 % (TT), un hi lu trong cch thy 3h v sau c
dch ny n cn. Ha tan cn trong 2 ml cloroform (TT).
Dung dch i chiu: Ha tan sarsasapogenin chun trong cloroform
(TT) c dung dch cha 5 mg/ml lm dung dch i chiu. Nu
hng c sarsasapogenin, ly 5 g bt Tri mu (mu chun) ri chit
nh mu th.
Cch tin hnh: Chm ring bit ln bn mng 7 l mi dung dch
th v dung dch i chiu. Trin khai xong, ly bn mng ra phi
kh ngoi khng kh. Phun hn hp ca dung dch vanilin 8 % trong
ethanol khan (TT) v dug dch acid sulfuric (0.5:5) [pha dung dch
acid sulfuric: Ly 2 ml nc, thm cn thn 7 ml acid sulfuric (TT),
lm lnh, thm nc va 10 ml]. Sy bn mng 5 pht 100 oC.
Trn sc l ca dung dch th phi c cc vt cng mu sc v
gi tr Rf vi cc vt trn sc k ca dung dch i chiu.
