Standard Ammonium Solution: - Dilute 10-ml stock ammonium solution to 1000 ml


with distilled water.
1 ml = 10 ug N = 12.2 ug NH3
Procedure
Treatment of Undistilled Samples
1. If necessary, remove residual chlorine from the freshly collected sample by addition an
equivalent amount of dechlorinating agent.
2. Add 1-ml ZnSO4 solution to 100-ml sample and mix thoroughly.
3. Add 0.4 to 0.5 ml of 6N NaOH solution to obtain a pH of 10.5 as determined with a pH
meter.
4. Mix gently. Let treated sample stand for a few minutes, where upon a heavy floceulent
precipitate should fall, leaving a clear and colourless supernate.
5. Clarify by centrifuging or filtering.
6. Take 50 ml treated sample for develop the colour.
7. Add 0.05 ml EDTA reagent or 1 to 2 drops Rochelle salt solution.
8. Mix well.
9. Add 2.0-ml nessler reagent if EDTA reagent is used or 1.0 ml nessler reagent if Rochelle
salt is used.
10. Measure absorbance with spectrophotometer at 400 to 425 nm.
