Microbial analysis of water

1. Dispense 1ml of raw water and treated water sample into four petri dishes
respectively.
2. Add 15ml-20ml of molten (cooled about 45oc) Nutrient agar, MacConkey
agar, Salmonella Shigella agar and Sabouraud dextrose agar into each petri
dishes.
3. Cover the petri dishes, mix the raw water and treated water respectively with
the molten agar by tilting or rotating the dishes.
4. Allow the content to solidify at room temperature
5. Invert the petri dishes and incubate at 30-37oc for 3 days for bacterial count
and 25oc for 3 days for fungi count.
6. After incubation, examine the plates for growth, count the number of
colonies on each plate and express the average for the 3 days in terms of the
number of colony forming unit per ml.

7. Specification:

Bacteria count must be less than 100cfu/ml.


Fungi count must be less than 100cfu/ml.
Coliform organisms must be absent in 1ml.
