Application of Ion-Exchange Resins: The Separation of Cations

I. Objectives

After completing this experiment, the students should be able to:

Describe the process of ion exchange chromatography


Draw the schematic diagram of the apparatus used in the experiment
Perform the calculations involved ion exchange chromatography

Deionized Water HNO3


5% (v/v) HCl 6 M NH4OH
6 M HCl Eriochrome Black T
2 M HCl EDTA

IV. Procedure

A. Pre-experiment Preparation

Equipment set up:


Each lab set up has extra tubing on the inlet side. The extra tubing is made to go into the
solution barrel. There is a section of tubing on that extra tubing. The section of tubing is the
section of tube that must go into the pump. Both lab set ups are using the same pump head so if
the system should not have flow, the tubing must be removed from the pump head. The pump
should be between the two lab setups.
Fill a 2L beaker with Deionized Water and use it as the source for the backwashing. The
5% (v/v) HCl will be used during the regeneration. The lab will have pre-made hard water for
processing to deionize.
B. Experiment

1. A 50 cm3 of the unknown solution containing Mg and Zn was pipetted into a clean 100
cm3 volumetric flask.

2. Then 25 cm3 of 6 M HCl was added and diluted with distilled water to the mark then well
mixed.
3. In 25 cm3 of the diluted mixture was pipetted onto the column. Then 250 cm3 conical
flask was placed beneath the column and the liquid was slowly drained until its level was
barely above the resin.
4. The Mg was eluted with about 50 cm3 of 2 M HCl at a flow rate of 3 cm3/min. The eluent
was collected.
5. The Zn (II) was eluted with 30 cm3 of water then 80 cm3 of HNO3 using the same flow
rate. The eluent was collected in 500 cm3 conical flask.
6. 10 cm3 of 6 M NH4OH was added to all collected eluate.5cm3 of buffer solution and 150
cm3 of water, then 2drops of Eriochrome Black T was added.
7. The solutions were titrated with standard EDTA until the color change from red to blue.
