Question 1:

Rheumatic heart disease (RHD) is the most serious irreversible complication of rheumatic fever
in many countries of the word. Repeated inflammation with fibrinous repair may result in
changes anatomical and function of affected valves. Since circulating endothelial cells (CEC)
have been identified as a surrogate marker of endothelial dysfunction, we therefore aimed to
determine the correlation of CECs and soluble vascular cell adhesion molecule-1 (VCAM-
1) with lesion severity in rheumatic heart disease (RHD) patients. A comparison study will
perform to compare the expression of CECs and VCAM-1 on the CECs among mild, moderate
and severe valve lesion Subjects will be 36 children aged 6 to 14 years, divided into two groups:
18 RHD patients and 18 healthy controls group respectively.

1. Please mention and explain the method that can be used to determine and measure the
CEC amount in this study and attach the kit/antibody data sheet.
2. Please mention and explain the method that can be used to determine and measure the
VCAM-1 expression in this study and attach the kit/antibody data sheet.
