Introduction: Cancer is a disease of growth and development disorders in the

abnormal cells of the body. Cancer will form new invasive or malignant tissues.
Numerous benefit of soursop as anti-cancer drug are making people to start
switching to soursop-leaf herb supplements as an alternative to conventional
preventions and treatments, but, it is not yet known how this herb works. This
study aims to determine the potential of essential oil and ethanolic extract of
soursop leaves as an anti-cancer through the activation of caspase 3 and Bax in
silico.
Method: The study was conducted by In silico with molecular docking method
about the active substance of essential oil and ethanolic extract of soursop
(Annona muricata Linn) leaf against the target protein Caspase 3 and Bax to
determine the values of the binding free energy, as well as hydrogen bonds and
hydrophobic bonds on the bond of amino acid residues.
Result: The active substance of essential oil and ethanolic extract of soursop
leaves have the potential to activate Caspase 3 with lower strength than
Doxorubicin drug control. The active substance of the essential oils β-caryopyllen,
Transcaryopyllen and β-cadinene have the ability to bind and activate lower Bax
than Doxorubicin. The active substance of Kaempferol 3-O-routoside and Routine
ethanolic extract have the same potential as Doxorubicin which can induce cell
apoptosis through intrinsic pathways.
Conclusion: The active substances in the essential oil and ethanolic extract can
activate Bax better rather than Caspase 3
Suggestion: To prove the results of computational prediction that has been done,
it is necessary to do further research using in vivo method and toxicity test to
prove the potential that essential oil and ethanol extract of soursop leaves as anti
cancer against activation activity of caspase 3 and Bax as proapoptosis protein.
