Tujuan Pewarnaan :
a. Mengetahui bahwa bakteri dapat dilihat dengan
pewarnaan
b. Mengenal bakteri positif Gram dan negatif Gram
serta morfologinya

1. Gelas alas bersih dan bebas lemak


2. Umur biakan (optimal) : 18 - 24 jam kecuali
bakteri tahan asam (M. tuberculosis)
3. Kualitas zat warna
4. Tebal tipisnya sediaan

Hasil pewarnaan :

bakteri Gram positif : ungu


bakteri Gram negatif : merah

Gently stain with crystal violet Gently stain wash off the stain Gently apply Gram’s iodine for 45 –

Counterstain with Safranin for 45

a. Kinyoun Gabbet (Tan Thiam Hok)


b. Ziehl Neelsen (WHO)

Prinsip :

a. Fukhsin karbol
 ikat (cuci dengan asam alkohol);
 tahan asam (merah)

b. Fukhsin karbol
 lepas (cuci dengan asam alkohol);
 bukan tahan asam (biru)
