IMMUNOENZIMACTIC ANALYSIS OF BONE MATRIX PROTEIN EXPRESSION IN PRE-OSTEOBLASTIC CELLS

INDUCED BY DIFFERENT METHODS OF PREPARATION HUMAN TEETH. IN-VITRO STUDY.

The similarity between the bone tissue and the dental tissue was the starting point for
the research in order to verify the osteoinductive potential of this tissue. Among the
studies the methods of preparation of the dental element and the use of graft material
are observed. In order to quantify the expression of type I collagen (COL-I), osteopontin
(OPN) and osteonectin (OCN) in cultures of pre-cultured cells, the in vitro study was
tested with several methods of tooth preparation - osteoblastics. In order to do this, 12
upper molar teeth were divided into four groups according to the preparation mode: FT
(full tooth) - teeth containing all tissues: enamel, dentin, pulp, cementum and
periodontal ligament (n = 3) . ); We (without enamel) - the teeth had the enamel
removed (n = 3); WPL (without periodontal ligament) - teeth with periodontal ligament
removal; and, WEPL (without enamel and periodontal ligament) - from the periodontal
ligament to the enamel. After the treat in groups, the teeth were triturated by
homogenous and then the extra pontluded proteins of the shredded teeth in incubated in
the cell of pre-osteoblastic women in 24h and 72h, in order to verify the potential
osteoinductive by means of immunoenzymatic analysis (ELISA). After analyzing the
data, it can be observed that COL-I was expressed differently between groups, without
statistical difference (p> 0.05). Regarding the OPN, it was the variation in the statis-
tics (p <0.05) between the FT and WEPL groups and a tendency for greater expression
in the FT and WE groups, but this was not statistically significant. The OCN did not
reach the expression in the times and in quantities studied. From the results obtained, it
can be concluded that the different methods of dental preparation tested are no longer
capable of osteoarthritis superior to the control group in relation to the expression of
collagen type I, osteopontin and osteocalcin.
