CONSTRUCTING MOLECULAR PHYLOGENY OF GENE nrLSU FOR

SUPPORTING THE IDENTIFICATION OF SAMPLE T004 AND T011


BELONGING TO THE PARASITIC CICADIDAE.

ABSTRACT
Cicadas (Family: Cicadidae) was known as the host of member
entomopathogenic fungal including 3 species of Metarhizium, 11 species
Ophiocordyceps, 5 species Polycephalomyces, .etc. distributed in the tropical
and subtropical region in Thailand, Southeast Asia (Prayook 2013), North
America (Mains 1939), and Europe (Kobayasi & Shimizu 1963). According to
the classification of Kobayashi (1982), specimen T004 and T011, are defined
respectively as Ophiocordyceps sobolifera and Cordyceps cicadae based on
their morphology, collected at Eakar district, Dak Lak province. The advent of
molecular biology, Large subunit (LSU) of rDNA (nrLSU) was supported the
described new species (Kumari et al., 2011, Shao et al., 2014), distinguished
the classification of Clavicipitaceae clade (Sung et al., 2007). In order to further
support the identification, we continued to analyze the nuclear ribosomal large
subunit gene region (nrLSU) on this specimen. The molecular phylogeny of the
nrLSU gene, analysis based on the Method for tree construction by a software
MEGA X, consisted of 79 sequences of Clavicipitaceae clade including 3
sequences of the outgroup. As a result, T004 was high supported as
Ophiocordyceps sobolifera, T011 was high supported as Cordyceps cicadae
(Anamorphic form: Isaria cicadae). The molecular phylogeny of nrLSU was
assistanced significant for identification of the fungal samples, collected at Eakar
district, Dak Lak province.
