This method depends its success upon the concentration and flotation of the cysts of the intestinal

amoebae and flagellates and is only applicable to the examination of formed stools, for the motile
trophozoites of these organisms would be destroyed during the process and these, as already noted
occur in fluid or semi-fluid stools. Material and reagents: Zn S04 33% (specific gravity of 1.180):

Technique: 1. Thoroughly mix one part of the formed stool, about the size of a pea.with about 10
part of a top water in clean glass container. Strain 10 cc of this mixture through on a layer of wet
cloth previously placed in a small funnel, into a tube centrifuge. The filtrate in the tube is then
centrifuge for forty-five to sixty seconds at the top speed of an International clinical centrifuge. The
supernatant fluid is poured off, 2 or 3 cc of distilled water added the tube thoroughly shaken to
distribute the sediment and additional water added to fill the tube. The tube is then centrifuge as
before and the process is repeated until the supernatant fluid is clear. After the last centrifugation,
the clear supernatant fluid is poured 1. m 4 off, and 3 to 4 cc of 33% zinc sulphate solution, which
should have a specific gravity of 1.180 are added. The sedimernt is thoroughly mixed with this
solution and enough of the zine sulphate solution added to fill the tube to within one-half inch of n
the rim. The tube is then centrifuge for at least ninety seconds at too speed 6. Several platinum
loopfuls of the material which will be noted floating upon the surface of the solution in the tube are
removed and placed upon a microscopic slide, one drop of D Antoni's iodine stain is added and
mixed. The preparation is covered 5 with a cover-slip and examined. Formalin-Ethyl Acetate or
