Special stains
also called histochemical stains, are use for detection specific substances or
constituents of the cells/tissues.
Aims of special stains
1- Help of diagnosis .
2- Reduce of time.
3- Perfect diagnosis for special substances.
4- Reduce the error in diagnosis .
5- To differentiate between constituents of tissue .
6- E.t.c
Sudan Black/Oil Red O
These stains are used for demonstration of Fat.
Sudan black and oil red O stains are use to stain of fat because these stains are more
soluble in fat . and use after Cut frozen section

Tissue with Oil Red O Stain

Fat : Bright red
Nuclei : Blue

Tissue with Sudan black stain

Fat : Black

Van Gieson
This stain is used for staining of collagen fibres.
Staining of collagen and other tissues (e.g. muscle) depending upon the
porosity of tissue and the size of the dye molecule.

Collagen: Red
Other tissues : Yellow
(including muscle
and RBC’s)

Masson’s Trichrome
This stain is used for staining of muscle.
Result
Muscle : Red
Collagen : Blue-green

Reticulin stain
This is used for demonstration of fibres.

Reticulin fibres : Black

Collagen: Brown

Congo Red
This stain is used for demonstration of Amyloid.

Amyloid elastic fibres : Red

Periodic Acid-Schiff (Pas)

PAS stain is used for demonstration of glycogen and mucopolysaccharides.

PAS positive substances : Magenta

Nuclei : Blue
PAS positive substances are glycogen, fungi,
basement membrane, neutral mucin and
hyaline cast.

Methyl Violet
It is used for demonstration of amyloid in tissue.

Metachromatic positive tissue : Red to violet

Other tissues : Blue

Prussian Blue/Perl’s Reaction

This is used for demonstration of Iron.

Iron : Blue
Cytoplasm and nuclei : Red to pink
