Staphylococcus

 Pigment products on potato agar (produces lighter color)


 Catalase products on trypticase soy agar (more opaque in color)

Potato agar = placed in a test tube

Slant- slanted position in test tube

Uninoculated means no bacteria yet

Inoculated- organism is placed

Uninoculated potato agar is yellowish-white

Catalase test is done on trypticase soy agar (TSA)

- prepared in a tube; also in a slant

- slanted position

- yellow if uninoculated

Hemolytic pattern and colonial morphology can be seen in blood agar plate

TSA yellow + human blood = malt red

Ideal red blood cells to be added on TSA blood agar is sheeps’ blood

- because it provides better results in hemolytic pattern

Mannitol Fermentation test is run always on Mannitol Salt Agar (MSA)

- color if uninoculated is faint orange

Best biological test = coagulase test= biochemical test that will differentiate Staphylococcus aureus from
Staphylococcus epidermidis

Representative organism for investigation

Catalase test- appearance of gas bubbles

Principle: test that will demonstrate the ability of organism to reduce hydrogen peroxide to water and
oxygen

Do not make conclusions right away.

Catalase = produced by organism an enzyme, never harmed by disinfectant

Rgt used: H2O

+ = appearance of gas bubbles not harmed by H2O2

3-5 loopfuls

After 24 hours, can identify colonial morphology

Colonies of Staph. Aureus

- smooth, mucoid, golden yellow colonies

- produces yellow pigment because of lipochrome

Best at room temperature for pigment of staph aureus

Staphylococcus epidermidis

- rough, round, white colonies

- produces white pigment (the striking feature between 2 staphs strain)

Blood Agar – an enrichment and differential media that supports growth of organism because it can
support all kinds of organisms

The growth could vary depending on the ability of the organism to utilize hemoglobin in red blood cells

Brown’s Classification

3 hemolysis plate

β/ beta hemolysis – complete destruction of RBC’s in blood agar plate
