PROCEDURE

Biuret’s Test Million’s Test

Mix 2ml of 10% NaOH Add 4 drops of mililon’s


with 2ml egg albumin reagent to 2ml of egg
albumin

Add 1 drop of 0.1%


CuSO and mix
Mix well, then warm the
solution by putting it to
water bath

If no pink or violet
appears, drop up to 10
of 0.1 CuSO4 then mix

Xanthoproteic Test

Add 1ml of
concentrated nitric Mix then warm the Carefully add
acid to 2ml of the mixture carefully concentrated HNO2
solution (albumin)

Hopkin’s-Cole Test

Incline the tube in


Add 2ml of Hopkin-Cole angle, Carefully add If there’s no
reagent to 2ml albumin 3ml of concentrated appearance of violet
H2SO4 ring, agitate the tube
Reduce Sulfur Test

Add 5drops of lead


acetate solution, boil
Add 2ml of 6M NaOH to Heat carefully in flame
again if there’s no
2ml albumin for 15 seconds
appearance of black
precipitate
