AGAR PLATES:-

a) Plug the Erlenmeyer flask with molten agar medium with caution.

b) Sterilize the flask of agar medium.

c) Remove the flask from autoclave and mix contents well with
vigorous swirling motions.

d) Cool these flasks and sterilize again in a water bath. This process is
called tempering the agar.

e) Remove the tempered agar from the water bath and wipe the
water of the flask.

f) Hold the neck of the Erlenmeyer flask with the hand you write
with.

g) Turn the other palm side up and clamp the cotton plug between
two fingers.

h) Remove the plug and flame the mouth of the flask.

g) Use the hand holding the cotton plug to tilt the lid of the petriplate.
