1. Cell cycle arrest pada G1 : siklus sel yg berhenti pd fase Growth pertama, dimana yg
seharusnya ada G(G1: ukuran sel Growth factor, struktur DNA G2: replikasi DNA),
S(sintesis) dan M (M1 M2 metosis meiosis.
Interfase trdapat di sebelum G1
Berhentinya siklus sel, dimana siklus sel terdiri atas 2 fase utama yaitu :
a. Fase S (Sintesis) : terjadi replikasi kromosom DNA
b. Fase M (Mitosis) : pemisahan 2 set DNA kromosom tersebut menjadi 2 sel
(Nurse, 2002).
a. Gap 1 : (sebelum fase S) persiapan untuk sintesis DNA yang merupakan fase awal
siklus sel
b. Gap 2 : (setelah fase S) el melakukan sintesis lebih lanjut untuk proses
pembelahan pada fase M (Ruddon, 2007).

Setelah fase M, berlanjut pada fase G1 atau G0.

Cells are constantly monitoring their cell cycle status through cell cycle checkpoints.
Cyclins and cyclin-dependent kinases regulate cell progression through the stages of the
cell cycle (G1, S, G2, M). Failure of the first two checkpoints (G1, G2/M) mostly occurs
upon DNA damage. The G1 checkpoint monitors the DNA integrity and cell machinery
and determines whether the cell will undergo DNA replication (S-phase) [1]. Cells that
fail the G1 checkpoint due to DNA damage can initiate a process of DNA repair or
initiate apoptosis. DNA damage can cause cell cycle arrest through induction of the p53
pathway, which can lead to initiation of repair or apoptosis[2]. Cells that fail the G1
checkpoint due to limited nutrients or extracellular signaling can enter the G0 phase of
the cell cycle, which describes a state of quiescence. Many cells of the body are stuck in
a permanent state of cell cycle arrest (i.e. terminally differentiated) and are unable to
replicate (e.g. neurons, muscle cells). However, under a specific set of conditions,
muscle cells can enter mitosis if stimulated by growth factors or by the expression of a
retroviral oncogene [3].
After cells pass the G1 checkpoint, they can progress through the initial stages of DNA
replication during S phase, and they must progress through the entirety of S phase to G2
phase before encountering the next checkpoint, the G2/M checkpoint. DNA damage can
arrest cells in G2 before progressing to mitosis, and in G2, the cell can undergo DNA
repair mechanisms or apoptosis similarly to during G1. If the cell passes the G2/M
checkpoint, cells begin to undergo mitosis until the spindle checkpoint. The cell will not
progress through mitosis until the spindles align at the equator of the cell. This process
is incredibly important because it ensures that both daughter cells receive one
chromatid from each pair of sister chromatids. Cells that fail this checkpoint become
arrested in anaphase [4]. After this checkpoint, cells can finish mitosis and cytokinesis to
generate two daughter cells.

4 jenis design penelitian:
- pre experiment design bukan merupakan exp sungguh2 krn ada factor luar yg
mmpengaruhi variable dependent. Masih banyak perancu nya dan blm focus pd titik
yg dicari.
Bentuk design: one shoot case study, one group pretest post test design, intake
group comparation.
- True experiment design design dpt mngontrol semua variable dari luar,
validitasnya bisa dipercaya dari pda pre experimental, sample nya dipilih scr random,
memiliki 3 design penelitian: post tes only, pre post test, Solomon-four group
- Quasi experiment design merupakan pengmbangan true experiment design, sulit
dilaksanakan punya kelompok kontrol tp tdk berfungsi untuk mengotrol variabel2
luar. Lebih valid dibandingkan pre experimental design, model desing : Time series,
non equivalen control group design, conter balances design
- Factorial design selalu melibatkan 2 atau lebih variable bebas, tujuan nya untuk
mengetahui apakah efek dpt digeneralisasikan lewat semua level dari semua
variable control.

There are three broad categories of experiments: in vitro studies, in vivo studies, and in silico
studies. Each study type has conveniences and liabilities. Understanding the liabilities of study
types offers insight into the validity of researchers' conclusions.
In vitro (Latin for within the glass) refers to the technique of performing a given procedure in a
controlled environment outside of a living organism. Many experiments in cellular biology are
conducted outside of organisms or cells. One of the abiding weaknesses of in vitro
experiments is that they fail to replicate the precise cellular conditions of an organism,
particularly a microbe. To cite one example among many, the lysates or extracts from culture-
grown spirochetes do not reflect antigens expressed in the mammalian Borrelia:
Addressing this question is complicated by the fact that protein expression of culture-grown
spirochetes does not fully resemble Borrelia in the host, i.e. the usefulness of protein lysates
from culture-grown bacteria is limited as a source of antigen for ELISPOT analysis.

Because of this, in vitro studies may lead to results that do not correspond to the
circumstances occuring around a living organism.
Until the last several years, efforts to detect and identify microorganisms in the human body have
depended almost exclusively on in vitro studies. As a result, many researchers began to assume
that chronic diseases were not caused by microbes. The net effect of all this was that the
understanding of pathogens in disease was driven by the study of well-known, easy-to-culture
microbes–which, as it turns out, represent the vast minority of bacteria in the human body. By
one estimate, 99.6% of the species in the human microbiota have not or cannot be characterized
through in vitro techniques.2)
In vivo studies
In vivo (Latin for “within the living”) refers to experimentation using a whole, living organism as
opposed to a partial or dead organism. Animal studies and clinical trials are two forms of in vivo
research. In vivo testing is often employed over in vitro because it is better suited for observing
the overall effects of an experiment on a living subject.
While there are many reasons to believe in vivo studies have the potential to offer conclusive
insights about the nature of medicine and disease, there is a number of ways that these
conclusions can be misleading. For example, a therapy can offer a short-term benefit, but a long-
term harm.
In silico studies
In silico is an expression used to mean “performed on computer or via computer simulation.”
The expression in silico was first used in public in 1989 in the workshop “Cellular Automata:
Theory and Applications” in Los Alamos, New Mexico. Pedro Miramontes, a mathematician
from National Autonomous University of Mexico (UNAM), presented the report “DNA and
RNA Physicochemical Constraints, Cellular Automata and Molecular Evolution.” In his talk,
Miramontes used the term “in silico” to characterize biological experiments carried out entirely
in a computer.
Although in silico studies represent a relatively new avenue of inquiry, it has begun to be used
widely in studies which predict how drugs interact with the body and with pathogens. For
example, a 2009 study used software emulations to predict how certain drugs already on the
market could treat multiple-drug-resistant and extensively drug-resistant strains of tuberculosis.3)
There is a variety of in silico techniques, but the two that are discussed the most in connection
with the Marshall Protocol are:
Bacterial sequencing techniques – As an alternative to in vitro methods for identifying bacteria,
various in silico methods which sequence bacterial DNA and RNA have been developed. The
most commonly used use is polymerase chain reaction (PCR). PCR takes a single or few copies of
a piece of DNA and increases it across several orders of magnitude, generating millions or more
copies of a particular DNA sequence. PCR has allowed researchers to detect bacteria associated
with a variety of conditions with increasingly high sensitivity.
Molecular modeling – Part of the Marshall Pathogenesis is based on in silico work,
demonstrating how drugs and other substances interact with the nuclear receptors of cells. In
particular, Trevor Marshall, PhD, has used computer-based emulations to show that 25-D, one
of the vitamin D metabolites, and Capnine, a substance produced by bacteria, turn off the
Vitamin D Receptor. These conclusions have since been validated by clinical observations.
Whole cell simulations – As described here, researchers have built a computer model of the
crowded interior of a bacterial cell that—in a test of its response to sugar in its environment—
accurately simulates the behavior of living cells.4)

Dalam penelitian in vitro yang lebih cocok dibandingkan in vivo untuk menyimpulkan
tindakan mekanisme biologis. Dengan variabel yang lebih sedikit dan perseptual
diperkuat menyebabkan reaksi halus, hasil yang umumnya lebih jelas.

- uji pada mikroba jika antibiotic;
- pada sel kanker dari hewan utk obat anti kanker;
- pada plasmodium utk obat anti malaria;
- pada jamur missal candida pada obat anti keputihan/candidiasis;
- pada cacing utk obat cacing;
- pada virus utk obat antivirus;
- pada bagian organ tertentu dari hewan contoh obat asma bronkodilator diuji pada
otot polos trachea marmot;
- pada jantung hewan dalam chamber utk obat angina dan aritmia; dll.
kekurangan in vitro :
- Banyak percobaan biologi seluler dilakukan di luar organisme atau sel ; karena
kondisi pengujian mungkin tidak sesuai dengan kondisi di dalam organisme, ini dapat
mengakibatkan hasil yang tidak sesuai dengan situasi yang muncul dalam
organisme hidup. Akibatnya, hasil eksperimen tersebut sering dijelaskan dengan in
vitro, bertentangan dengan in vivo.
- Namun, kondisi yang terkendali hadir dalam sistem in vitro berbeda secara signifikan
dari yang in vivo, dan dapat memberikan hasil yang menyesatkan. Oleh karena itu,
dalam studi in vitro biasanya diikuti oleh studi vivo.
Contohnya termasuk:
- Dalam biokimia, fisiologis stoikiometri konsentrasi non-aktif dapat mengakibatkan
enzim dalam arah terbalik, misalnya beberapa enzim dalam siklus Krebs mungkin
tampak memiliki tata-nama, salah.
- DNA dapat mengadopsi konfigurasi lainnya, seperti A DNA .
- Protein lipat mungkin berbeda seperti dalam sel ada kepadatan tinggi protein lain
dan ada sistem untuk membantu lipat, sementara in vitro, kondisi kurang
bergerombol dan tidak membantu.

Syarat hewan yg digunakan sangat banyak tgt jenis obatnya, missal yang jelas harus
dilakukan control terhadap galur/spesies, jenis kelamin, umur, berat badan
(mempengaruhi dosis)
harus dilakukan pada minimal 2 spesies yakni rodent/hewan mengerat dan non
rodent. Alasannya krn system fisiologi dan patologi pada manusia merupakan
perpaduan antara rodent dan non rodent.

2. Faktor eksternal
Meliputi suplai oksigen, pemeliharaan lingkungan fisiologik (keadaan
kandang, suasana asing atau baru, pengalaman hewan dalam
penerimaan obat, keadaan ruangan tempat hidup seperti suhu,
kelembaban, ventilasai, cahaya, kebisingan serta penempatan hewan),
pemilihan keutuhan struktur ketika menyiapkan jaringan atau organ
untuk percobaan. Faktor-faktor tersebut dapat mempengaruhi hasil
percobaan, dan mempengaruhi efek farmakologinya, apabila hewan
yang sudah biasa di beri obat maka akan terlihat lebih rilex dan santai
berbeda dengan hewan percobaan yang masih baru dan masih asing
makan akan lebih berontak dan agresif, sehingga kita membutuhkan
penelitian dan perawatan yang baik terhadap hewan percobaan
sebelum melakukan percobaan.

Spesies yang ideal untuk uji toksisitas sebaiknya memenuhi criteria-kriteria sebagai berikut:
Berat badan lebih kecil dari 1 kg
Mudah di ambil darahnya dan jumlah darah yang dapat diambil cukup banyak
Mudah dipegang dan dikendalikan
Pemberian materi mudah dilakukan dengan berbagai rute
Mudah dikembangbiakan dan mudah dipelihara di laboratorium
Lama hidup relative singkat
Fisiologi diperkirakan sesuai/identik dengan manusia/hewan yang dituju

Penelitian yang memanfaatkan hewan coba, harus menggunakan hewan percobaan yang
sehat dan berkualitas sesuai dengan materi penelitian. Hewan tersebut dikembang-biakkan
dan dipelihara secara khusus dalam lingkungan yang diawasi dan dikontrol dengan ketat.
Tujuannya adalah untuk mendapatkan defined laboratory animals sehingga sifat genotipe,
fenotipe (efek maternal), dan sifat dramatipe (efek lingkungan terhadap fenotipe) menjadi
konstan. Hal itu diperlukan agar penelitian bersifat reproducible, yaitu memberikan hasil
yang sama apabila diulangi pada waktu lain, bahkan oleh peneliti lain.
Berbagai hewan kecil memiliki karakteristik tertentu yang relatif serupa dengan manusia,
sementara hewan lainnya mempunyai kesamaan dengan aspek fisiologis metabolis
manusia.
Dalam penelitian kesehatan yang memanfaatkan hewan coba, juga harus diterapkan prinsip
3 R dalam protokol penelitian, yaitu: replacement, reduction,dan refinement.
Replacement adalah keperluan memanfaatkan hewan percobaan sudah diperhitungkan
secara seksama, baik dari pengalaman terdahulu maupun literatur untuk menjawab
pertanyaan penelitian dan tidak dapat digantikan oleh mahluk hidup lain seperti sel atau
biakan jaringan. Replacement terbagi menjadi dua bagian, yaitu: relatif (mengganti hewan
percobaan dengan memakai organ/jaringan hewan dari rumah potong, hewan dari ordo
lebih rendah) dan absolut (mengganti hewan percobaan dengan kultur sel, jaringan, atau
program komputer).
Reduction diartikan sebagai pemanfaatan hewan dalam penelitian sesedikit mungkin, tetapi
tetap mendapatkan hasil yang optimal. Jumlah minimum biasa dihitung menggunakan
rumus Frederer yaitu (n-1) (t-1) >15, dengan n adalah jumlah hewan yang diperlukan dan t
adalah jumlah kelompok perlakuan. Kelemahan dari rumus itu adalah semakin sedikit
kelompok penelitian, semakin banyakjumlah hewan yang diperlukan, serta sebaliknya.
Untuk mengatasinya, diperlukan penggunaan desain statistik yang tepat agar didapatkan
hasil penelitian yang sahih.
