    gaino 4oIND Forensic DNA Fingerprinting Kit Quick Guide 7 Lesson 1 Restriction Digestion 1. Place the tube contalning the restc- ‘ion enzyme mi, labeled ENZ, once. 2. Label one of each colored micro test ‘tubes as follows: ‘green tube CS (crime scene) blue ube St (suspect 1) ‘orange tube $2 (suspect 2) violettube $3 (suspect 3) redtube $4 (euspect 4) yellow tube $5 (suspect §) Labo the tubes with your name, date, ‘and lab period. Place the tubes in the foam micro test tube holder. 3. Using a fresh tip for each sample, pipet 10 ul of each DNA sample from the stock tubes and transfer to the corresponding colored micro test tubes. Make sure the sample is transfered othe botom of the tubes. 4. Pipot 10 pl of enzyme mix (ENZ) into the very botiom of each tube. Use a fresh tip to transfer the ENZ sample to each tube. Pipet up and down carefully to mic wel. 8. Tightly cap the tubes and mix the components by gently flicking the tubes with your finger. If a ricrocentrituge is available, pulse- spin in the centituge to collect al he liquid in the bottom of the tube. Otherwise, gently tap the tube on the table top. 16. Pace the tubes nthe foam miro tube holder ard incubate for 45 min at 37°C ‘or overight at room temperature in @ large volume of water heated to 37°C. 7. It required, follow the instructors
